Leicestershire Police was called to reports of the crash on the southbound carriageway of the A46, just before the Anstey junction, at about 18:00 GMT on Tuesday.
A man in his 20s, who was driving a Land Rover Defender, was pronounced dead at the scene while another man, in his 30s, was taken to hospital where he is being treated for life-threatening injuries.
A 53-year-old man, who was arrested on suspicion of causing death by dangerous driving, has since been released under investigation.
The A46 was closed but reopened on Wednesday after repair work was completed.
Detectives said the crash involved a lorry, a grey Land Rover Defender, a black Kia Ceed, a blue Volkswagen Sharan and a Volkswagen Transporter.
They are appealing for anyone who was driving in the area at the time to get in touch, particularly anyone with any dashcam footage.
